临床皮肤科杂志2025年54卷第10期JClinDermatol,October2025,Vol.54,No.10外用马尾松针提取物对双氢睾酮抑制的C57BL/6小鼠毛发生长的影响597论著朱红柳,高以红,毛秋霞,封乐池,徐白燕,羊剑秋江阴市中医院皮肤科,江苏 江阴214400[摘要】目的:观察外用马尾松针提取物(PMNE)对双氢睾酮(DHT)抑制的C57BL/6小鼠毛发生长的影响。方法:采用皮下注射DHT建立雄激素性秃发小鼠模型。将48只雄性C57BL/6小鼠随机分为空白对照组、DHT组、米诺地尔组及PMNE低、中、高浓度组(每组各8只)。空白组不予任何处理;DHT组仅予皮下注射DHT(每只30mg/d分区注射);米诺地尔组予皮下注射DHT联合外用2%米诺地尔(每日2次,每次0.2mL);PM NE低、中、高浓度组予皮下注射DHT联合外用PMNE(浓度分别为5%、10%、20%,每日2次,每次0.2mL),连续用药4周。28d后取小鼠背部圆形皮片,刮取毛发称重,进行苏木精-伊红(HE)染色,观察毛囊形态及大。⒎直鸺扑闵て、休止期毛囊数量及二者的比值。取小鼠部分背部皮肤组织,采用实时荧光定量聚合酶链式反应(qPCR)和Westernblot 检测白细胞介素(IL)-6、转化生长因子(TCF)-β2、肿瘤坏死因子(TNF)-αmRNA及其蛋白的表达水平。结果:与DHT组相比,PMNE能够呈浓度依赖性地诱导毛囊进入生长期,表现为随着PMNE给药浓度的增加,小鼠毛发质量、生长期毛囊数量及生长期/休止期毛囊比值均逐渐提高。PMNE呈浓度依赖性地抑制IL-6、T CF-β2、T NF-αmRNA及其蛋白的表达。结论:外用PMNE能够显著抑制DHT处理的 C57BL/6小鼠局部皮肤组织中 IL-6、T G F-β2、T NF-α等炎症因子的表达,有效促进毛发生长。[关键词]马尾松针提取物;雄激素性秃发;米诺地尔;白细胞介素-6;转化生长因子-β2;肿瘤坏死因子-α[中图分类号] R751doi: 10.16761/j.cnki.1000-4963.2025.10.006Effects of topical application of pinus massoniana needle extract on hair growth in C57BL/6 micetreated with dihydrotestosteroneZHU Hongliu, GAO Yihong, MAO Qiuxia, FENG Lechi, XU Baiyan, YANG JianqiuDepartment of Dermatology, Jiangyin Hospital of Traditional Chinese Medicine, Jiangyin 214400, China[Abstract] Objective: To study the effects of topical application of pinus massoniana needle extract(PMNE) on hair growth inC57BL/6 mice treated with dihydrotestosterone(DHT). Methods: Forty-eight male C57BL/6 mice were randomly divided into 6 groups:control group without any treatment,DHT group(30 mg/d DHT only),minoxidil-treated group(DHT+2% minoxidil solution), PMNElow dose group(DHT+5%PMNE), PMNE medium dose group(DHT+10%PMNE), and PMNE high dose group(DHT+20% PMNE). Histo-morphometric analysis was employed to study induction of the hair follicle cycle after PMNE and minoxidil were topically appliedto the shaved dorsal skin of telogen mice 0.2 mL, twice a day, for four weeks. Photography of the areas was done weekly. Regrownhairs were plucked from the central dorsal area of sacrificed mice after 28 days. The hair weight was measured. Some skin tissue wasobtained for H&E stain; the numbers and morphology of the hair follicles were studied. The level of mRNA and protein expressionof IL-6,TGF-β2, TNF-α in skin tissue were determined by quantitative real-time PCR(qPCR) and western-blot, respectively. Re-sults: Compared with the DHT group, PMNE promoted hair follicle to enter the anagen phase in a dose-dependent fashion, as ev-idenced by an increase in hair weight, the number of anagen follicles, as well as the ratio of the number of anagen follicles to thenumber of telogen follicles on H&E-stained sections. In addition, PMNE inhibited the expression of mRNA and protein of IL-6, TGF-β2and TNF-α in dose-dependent fashion.
