269德国洋甘菊精油对咪喹莫特诱导的BALB/c小鼠皮肤银屑病样损害的作用研究临床皮肤科杂志2025年54卷第5期JClinDermatol,May2025,Vol.54,No.5论著吕曹华1,廖国艳²,葛维维1,吕荫宜²,吴双²,杜云婷,陈光²1台州市第二人民医院皮肤科,浙江台州318000;2台州学院医学院病原生物与免疫学教研室,浙江台州318000;3辽宁省肿瘤医院检验科,辽宁沈阳110042[摘要】目的:研究德国洋甘菊精油(MCEO)预处理对咪喹莫特(imiquimod,IMQ)诱导的BALB/c小鼠银屑病样皮肤炎症的保护作用。方法:取28只6~8周体重约为20g的雌性BALB/c小鼠,随机分为3组:正常对照组(n=8)、IM Q 模型组(n=12)和MCEO预处理+ IMQ组(n=8)。正常对照组小鼠予橄榄油外涂背部皮肤,每日2次,连续7日;IMQ模型组小鼠予IMQ外涂背部皮肤,每日1次,连续7日;MCEO预处理+ IMQ组小鼠予MCEO外涂背部皮肤预处理,每日2次,连续7日,随后予IMQ(每日1次)+ MCEO(每日2次)连续外涂背部7日。每日观察小鼠皮损情况,根据银屑病皮损面积及严重程度指数(psoriasisareaand severity index,PASI)进行评分,HE染色观察皮肤损伤情况。采用实时荧光定量逆转录聚合酶链式反应(quantitative real-timereverse transcriptase polymerase chain reaction,qRT-PCR)检测小鼠皮肤组织中炎性细胞因子白细胞介素(IL)-6和IL-1βmRNA的表达水平,采用液相芯片技术(Luminex)检测小鼠外周血中炎性细胞因子IL-1β、肿瘤坏死因子(TNF)-α、干扰素(IFN)-、IL-10和IL-17A的水平。Western blotting法观察小鼠皮肤组织中丝裂原活化蛋白激酶 p38(p38MAPK)、磷脂酰肌醇3-激酶(PI3K)和哺乳动物雷帕霉素靶蛋白(mTOR)信号通路蛋白的表达水平。结果:与正常对照组相比,IMQ模型组小鼠背部出现明显的红斑、鳞屑和银屑病样皮损,表皮角化过度伴角化不全,炎性细胞浸润。与IMQ模型组相比,MCEO预处理+ IMQ组中MCEO预处理明显改善了小鼠皮损情况,角化过度和角化不全明显减轻,仅见少量炎性细胞浸润;小鼠皮肤组织中IL-6和IL-1βmRNA以及外周血中IL-1β、T NF-α和 IFN-的表达水平均显著降低(P均<0.05);小鼠皮肤组织中p-PI3K/PI3K、p-mTOR/mTOR和p-p38/p38MAPK蛋白的表达水平均显著降低(P均< 0.05)。结论:MCEO预处理可明显改善IMQ诱导的小鼠皮肤银屑病样损害及皮肤炎性细胞浸润,其保护性作用机制与MCEO预处理抑制PI3K/mTOR和p38MAPK信号通路活性、下调皮肤组织和外周血中 IL-6、IL-1β、T NF-α和IFN-等炎性细胞因子的表达水平密切相关。[关键词」德国洋甘菊精油;预处理;银屑。贿溧兀籅ALB/c小鼠;炎症[中图分类号]R758.63doi 10.16761/j.cnki.1000-4963.2025.05.003Effects of Matricaria chamomilla essential oil on psoriatic lesionsinduced by imiquimod in BALB/c miceLV Caohua', LIAO Guoyan’, GE Weiwei', LV Yinyi?, WU Shuang, DU Yunting, CHEN Guang?I Department of Dermatology, Taizhou Second people's hospital, Taizhou 318000, China; 2 Department of Pathogenic Biologyand Immunology, Medical college of Taizhou University, Taizhou 318000, China; 3 Department of Laboratory Medicine, Liaon-ing Cancer Hospital Insitute, Shenyang 110042, China[Abstract] Objective: To investigate the protective effects of Matricaria chamomilla essential oil(MCEO) on imiquimod(IMQ)pretreatment-induced psoriasis-like skin inflammation in BALB/e mice. Methods: The animals were divided into three groups:normal control(n=8), IMQ model group(n=12) and MCEO+IMQ group(n=8). The normal control group was treated with oliveoil applied externally to the back skin twice a day for 7 days.The BALB/c mice in IMQ model group were smeared withIMQ on their back skin once a day for 7 days. Firstly, the BALB/e mice in the MCEO+IMQ group were pretreated with MCEOfor 7 days and applied to back skin once in the morning and once in the evening, and then both IMQ(once daily) and MCEO(twice daily) were applied continuously for 7 days. Skin lesions in the mice were observed daily. Psoriasis area and severityindex(PASI) score and HE staining were used to confirm skin lesions. The expression levels of inflammatory cytokines IL-6and IL-1β mRNAs in skin tissues were detected by quantitative real-time reverse transcriptase polymerase chain reaction(qRT-PCR). The levels of inflammatory cytokines IL-1β, TNF-α, IFN-, IL-10 and IL-17A in the peripheral blood of micewere measured by liquid phase chip technology(Luminex). The protein levels of p38 MAPK, PI3K and mTOR signaling path-way were observed by western blotting. Results: Compared with the normal control group, the back skin in IMQ model groupshowed obvious erythema, scale and psoriatoria-like lesions, epidermal hyperkeratosis with hypokeratosis, inflammatory cell infiltration.
