Methods: Skin BCC and adjacent normal tissues were col-lected from surgical specimens in our hospital from January 2020 to December 2023, and the relative mRNA and protein ex-pression levels of TBX1 were detected. Skin BCC cell line TE354T was cultured, grouped and transfected with negative con-trol(NC), small interfering RNA(siRNA), TBX1 siRNA, blank vector, Sonic hedgehog(Shh) overexpression vector, Gli familyzinc finger(Gli) 2 overexpression vector, respectively. Then, the cells were treated with different concentrations of Shh in-hibitors(1 μmol/L, 5 μmol/L, 10 μmol/L) and Gli2 inhibitors(5 μmol/L, 10 μmol/L, 20 μmol/L). The absorbance value(A) ofcell proliferation, apoptosis rate, and relative mRNA and protein expression levels of TBX1, Sh, Glil and Gli2 were detect-ed. Results: The relative mRNA and protein expression levels of TBX1 in skin BCC tissues were higher than those in nor-mal skin tissues(P<0.05). Compared with the si-NC group, the A values at 24 h, 48h and 72h and the relative expressionlevels of TBX1, Shh and Gli2 in TE354T cells in the si-TBX1 group were decreased(P<0.05). The cell apoptosis rate in thesi-TBX1 group was increased(P<0.05), while the relative expression level of Glil had no significant diference(P>0.05). TheA values at 24h, 48h and 72h of TE354T cells in the Shh inhibitor group and Gli2 inhibitor group were significantly de-creased(P<0.05). Compared with the si-TBX1+blank plasmid group, the A values at 24 h, 48 h and 72 h of TE354T cellsin the si-TBX1+Shh plasmid group and the si-TBX1+Gli2 plasmid group increased, and the cell apoptosis rate were decreased(P<0.05).
