However, bovine herpes-virus 1 (BoHV1) and related ruminant alphaherpesviruseshave been reported to cross the species barrier. Bubalineherpesvirus 1 (BuHV1) is an alphaherpesvirus closelyrelated to BoHV1 and BoHV5. According to the serolog-ical cross-relationships between ruminant alphaherpesvi-ruses, several surveys have studied the occurrence ofBoHV1-related virus infection in wild and domesticruminant species. Recent studies in Argentina showed anincrease in serological prevalence against BoHV1 relatedviruses in water buffaloes (Bubalus bubalis) population.The aim of this study was to investigate the presence ofrelated ruminant alphaherpesvirus in the Argentinean waterbuffalo population. BuHV1 was successfully isolated from5 out of 225 buffaloes analyzed. One isolate was obtainedfrom nasal secretions, and the others were from vaginalswabs. The buffaloes belonged to four different farmslocated in northeastern Argentina. The isolates were char-acterized as alphaherpesvirus by direct immunofluores-cence using FITC-anti-BoHV1 IgG. Restriction analysisperformed with BamHI and BstEII on the complete genomeshowed differences between the isolates and those fromBoHV1 and BoHV5 subtypes. Phylogenetic analysis onboth UL27 and US6 showed similarity in tree topology.While three of the isolates grouped together with sequencesof BoHV5, two other isolates clustered separately.
